CD11c Antibody (404-0114-82) in Flow
BALB/c mouse splenocytes were stained with CD45R (B220) Monoclonal Antibody, eFluor 450 (Product # 48-0452-82) and 0.25 µg of Armenian Hamster IgG Isotype Control, Brilliant Violet 421 (Product # 404-4888-81) (left) or 0.25 µg of CD11c Monoclonal Antibody, Brilliant Violet 421 (right). Total viable cells were used for analysis, as determined by 7-AAD (Product # 00-6993-50).

Description: The N418 monoclonal antibody reacts with mouse CD11c, the integrin alphaX. CD11c non-covalently associates with beta2 integrin to form the CD11c/CD18 heterodimer. CD11c is expressed by dendritic cells, a subset of Intestinal Intraepithelial Lymphocytes (IEL) and some activated T cells. CD11c/CD18 binds to CD54, iC3b and fibrinogen and plays a role in leukocyte adhesive interactions. N418 binds to CD11c on splenic dendritic cells in the T-dependent areas of mouse spleen and precipitates a 150, 90 kDa heterodimer.
Applications Reported: This N418 antibody has been reported for use in flow cytometric analysis.
Applications Tested: This N418 antibody has been tested by flow cytometric analysis of mouse bone marrow cells. This may be used at less than or equal to 0.5 µg per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.
Brilliant Violet™ 421 (BV421) is a dye that emits at 423 nm and is intended for use on cytometers equipped with a violet (405 nm) laser. Please make sure that your instrument is capable of detecting this fluorochrome.
When using two or more Super Bright, Brilliant Violet™, Brilliant Ultra Violet™, or other polymer dye-conjugated antibodies in a staining panel, it is recommended to use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize any non-specific polymer interactions. Please refer to the datasheet for Super Bright Staining Buffer or Brilliant Stain Buffer for more information.

CD11, along with CD18, form a heterodimer adhesion molecule. In particular, CD11 is composed of CD11a, CD11b and CD11c. CD11a is a leukocyte marker that is expressed in B and T lymphocytes, macrophages, monocytes, neutrophils, basophils and eosinophils. CD11b is primarily expressed by monocytes, macrophages, natural killer cells, some B and T-cells, and granulocytes. CD11c is expressed in monocytes, macrophages, natural killer cells, some granulocytes and less so in a subset of lymphocytes.
