Caspase-1, also known as Interleukin-1β-converting enzyme (ICE), is an inflammatory caspase responsible for processing pro-inflammatory cytokines like IL-1 beta and IL-18, and inducing pyroptosis, a form of programmed cell death. The active form of caspase-1 is a heterotetramer consisting of two p20 and two p10 subunits. The p10 subunit arises from the cleavage of the pro-caspase-1 precursor, which is essential for the enzyme's activation and function. The p12 subunit is a fragment derived from the cleavage of the proenzyme, which is involved in enzyme regulation. Upon activation, caspase-1 cleaves its substrates, leading to the initiation of inflammatory responses and pyroptotic pathways. The regulation of these subunits and their precise activities are crucial for maintaining immune homeostasis and responding to infections and tissue damage.
