Dynamin 2 Antibody in Immunocytochemistry (ICC/IF) — Immunofluorescent staining of HeLa cells using Product # PA5-19800, anti-Dynamin 2 antibody. The cells were fixed with PFA (4%) for 10 minutes, permabilised with BSA (1%), normal goat serum (10%) and glycine (0.3 M) in 0.1% T-BST for 1 hour and exposed to the primary antibody at a concentration of 1 µg/mL overnight at 4C. The secondary antibody was a 448 fluorescence conjugated Goat anti-rabbit IgG (green) at a dilution of 1:1000. A WGA- 594 fluorescent conjugated stain was used to label plasma membranes (red) and the nuclei stain was DAPI (blue).

Dynamin 2 Antibody in Western Blot (WB) — Western blot was performed using Anti-Dynamin 2 Polyclonal Antibody (Product # PA5-19800) and a 110kDa band corresponding to Dynamin 2 was observed across cell lines and tissues tested. Whole cell extracts (30 µg lysate) of HeLa (Lane 1), SH-SY5Y (Lane 2), HEK-293 (Lane 3), MCF7 (Lane 4), K-562 (Lane 5), Hep G2 (Lane 6), Raji (Lane 7), Mouse Lung (Lane 8) and Rat Brain (Lane 9) were electrophoresed using Novex® NuPAGE® 4-12 % Bis-Tris gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1ug/ml) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L), Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).

Dynamin 2 Antibody in Western Blot (WB) — Western blot analysis of mouse Testis Tissue Lysate using Product # PA5-19800, Dynamin 2 primary antibody at a dilution of 1 µg/mL (lane 1). Staining of SK N BE Whole Cell Lysate at a dilution of 1 µg/mL (Lane 2). Staining of Y79 Whole Cell Lysate at a dilution of 1 µg/mL (lane 3). Blot treated with a secondary HRP-conjugated Goat polyclonal anti-Rabbit antibody was used at a dilution of 1:3000.

Dynamins represent one of the subfamilies of GTP-binding proteins. These proteins share considerable sequence similarity over the N-terminal portion of the molecule, which contains the GTPase domain. Dynamins are associated with microtubules. They have been implicated in cell processes such as endocytosis and cell motility, and in alterations of the membrane that accompany certain activities such as bone resorption by osteoclasts. Dynamins bind many proteins that bind actin and other cytoskeletal proteins. Dynamins can also self-assemble, a process that stimulates GTPase activity. Five alternatively spliced transcripts encoding different proteins have been described. Additional alternatively spliced transcripts may exist, but their full-length nature has not been determined.
