FASN Antibody in Western Blot (WB) — All lanes: Western blot analysis of Fatty Acid Synthase with FASN Polyclonal Antibody (Product # PA5-120019) at 1:1,000 dilution. Lane 1: Wild-type Hela whole cell lysate. Lane 2: FASN knockdown Hela whole cell lysate. FASN Polyclonal Antibody was shown to specifically react with FASN in wild-type Hela cells. Weakened band was observed when FASN knockdown samples were tested. Wild-type and FASN knockdown samples were subjected to SDS-PAGE. Proteins were transferred to a PVDF membrane and blocked with 5% NFDM in TBST for 1 hour at room temperature. The primary Anti-FASN antibody (1:1,000) and Anti-HSP90 antibody (1:10,000) were used in 5% BSA at room temperature for 2 hours. Goat Anti-Rabbit IgG H&L (HRP) Secondary Antibody at 1:200,000 dilution was used for 1 hour at room temperature.

Fatty acid synthase is a multifunctional protein. Its main function is to catalyze the synthesis of palmitate from acetyl-CoA and malonyl-CoA, in the presence of NADPH, into long-chain saturated fatty acids. In some cancer cell lines, this protein has been found to be fused with estrogen receptor-alpha (ER-alpha), in which the N-terminus of FAS is fused in-frame with the C-terminus of ER-alpha.
