FOXA2 Antibody in Immunocytochemistry (ICC/IF) — Immunofluorescence analysis of FOXA2 in Hep G2 cells. Samples were incubated with FOXA2 Monoclonal antibody (Product # MA5-42678) using a dilution of 1:100 (40x lens). Blue: DAPI for nuclear staining.

FOXA2 Antibody in Western Blot (WB) — Western blot analysis of FOXA2 in extracts of various cell lines. Samples were incubated with FOXA2 Monoclonal antibody (Product # MA5-42678) using a dilution of 1:1,000, followed by HRP Goat Anti-Rabbit IgG (H+L) at a dilution of 1:10,000. Lysates/proteins: 25 µg per lane. Blocking buffer: 3% nonfat dry milk in TBST. Detection: ECL Basic Kit. Exposure time: 3 min.

FOXA2 Antibody in Western Blot (WB) — Knockout of FOXA2 was achieved by CRISPR-Cas9 genome editing using LentiArray™ Lentiviral sgRNA (Product # A32042, Assay ID CRISPR655639_LV) and LentiArray Cas9 Lentivirus (Product # A32064). Western blot analysis of FOXA2 was performed by loading 60 µg of PANC-1 wild type (Lane 1), PANC-1 Cas9 (Lane 2) and PANC-1 FOXA2 KO (Lane 3) whole cell extracts. The samples were electrophoresed using NuPAGE™ 3-8% Tris-Acetate Protein Gel (Product # EA0378BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with FOXA2 Recombinant Rabbit Monoclonal Antibody (9O4M2) (Product # MA5-42678, 1:1,000) and detected by Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:5,000) using the iBright™ FL1500 (Product # A44115). Chemiluminescent detection was performed using SuperSignal™ West Atto Ultimate Sensitivity Substrate (Product # A38556). Loss of signal upon CRISPR mediated knockout (KO) using the LentiArray™ CRISPR product line confirms that antibody is specific to FOXA2.

FOXA2 Antibody in Immunoprecipitation (IP) — Immunoprecipitation of FOXA2 in 300 μg extracts of HepG2 cells. Samples were precipitated with 3 μg FOXA2 Monoclonal antibody (Product # MA5-42678). Western blot was performed from the immunoprecipitate using FOXA2 Monoclonal antibody (Product # MA5-42678) at a dilution of 1:1,000.

FOXA2 is a member of the forkhead class of DNA-binding proteins. These hepatocyte nuclear factors are transcriptional activators for liver-specific genes such as albumin and transthyretin. They also interact with chromatin. Similar family members in mice have roles in the regulation of metabolism and in the differentiation of the pancreas and liver. FOXA2 has been linked to sporadic cases of maturity-onset diabetes of the young. Transcript variants encoding different isoforms have been identified.
