Immunohistochemistry analysis of GAK in mouse testis tissue. The sample was formaldehyde fixed and a heat mediated antigen retrieval step in citrate buffer was performed. Samples were incubated with GAK polyclonal antibody (Product # PA5-99201) using a dilution of 1:100 (4°C overnight) followed by HRP conjugated anti-Rabbit secondary antibody.

Antibody detects endogenous levels of total GAK.

GAK serine/threonine kinase or Cyclin-G-associated kinase was originally identified as a protein that is associated with cyclin G. However, neither CDK5 nor GAK kinase activity is activated by cyclin G nor does GAK affect CDK5 kinase activity. GAK seems to act as an auxilin homolog that is involved in the uncoating of clathrin-coated vesicles by Hsc70 in non-neuronal cells. Expression oscillates slightly during the cell cycle, peaking at G1.
