Phospho-Smad3 (Ser425) Antibody in Immunocytochemistry (ICC/IF) — Immunocytochemistry analysis of Phospho-Smad3 (Ser425) in H2O2 treated Hela cells. Samples were incubated with Phospho-Smad3 (Ser425) polyclonal antibody (Product # PA5-104939) and mouse anti-beta tubulin antibody for 1 hour at 37°C. An AlexaFluor594 conjugated goat anti-rabbit IgG(H+L) (Red) and an AlexaFluor488 conjugated goat anti-mouse IgG(H+L) (Green) were used as the secondary antibodies. The nuclear counter stain is DAPI (blue).

Samples were fixed with paraformaldehyde, permeabilized with 0.1% Triton X-100, blocked with 10% serum (45 min at 25°C), incubated with mouse anti-beta tubulin and Phospho-Smad3 (Ser425) polyclonal antibody (Product # PA5-104939) using a dilution of 1:200 (1 hr, 37°C), and followed by goat anti-rabbit IgG Alexa Fluor 594 (red) and goat anti-mouse IgG Alexa Fluor 488 (green).

Antibody detects endogenous levels of Smad3 only when phosphorylated at Serine 425.

SMAD3 belongs to the SMAD family of proteins. They are similar to the gene products of the Drosophila gene 'mothers against decapentaplegic' (Mad) and the C. elegans gene Sma. SMAD proteins are signal transducers and transcriptional modulators that mediate multiple signaling pathways. This protein functions as a transcriptional modulator activated by transforming growth factor-beta and is thought to play a role in the regulation of carcinogenesis.
