A peroxidase-conjugated goat anti-mouse IgG at 1:400 dilution was used as the secondary antibody, followed by DAB staining.

RAC1 Antibody in Western Blot (WB) — Western blot was performed using RAC1 Monoclonal Antibody (1301CT276.121.104) (Product # MA5-37658) and a 21 kDa band corresponding to RAC1 was observed across all cell lines and tissues tested. Whole cell extracts (30 µg lysate) of HeLa (Lane 1), MCF7 (Lane 2), A-431 (Lane 3), PC-3 (Lane 4), A549 (Lane 5), HT-29 (Lane 6), NIH/3T3 (Lane 7), Mouse Brain (Lane 8) and Rat Brain (Lane 9) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0321BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1,000) and detected by chemiluminescence with Goat anti-Mouse IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177, 1:20,000) using the iBright™ FL1500 Imaging System (Product # A44115). Chemiluminescent detection was performed using SuperSignal™ West Pico PLUS Chemiluminescent Substrate (Product # 34580). An uncharacterized band at ~ 60 kDa was observed in the samples tested.

RAC1 Antibody in Western Blot (WB) — Western Blot analysis of RAC1 using the RAC1 Monoclonal Antibody (Product # MA5-37658) at a dilution of 1:1,000 in A431, mouse NIH/3T3, rat C6 cells (from left to right) (35 µg per lane). A goat anti-mouse IgG H&L(HRP) at 1:3000 dilution was used as the secondary antibody.

RAC1 Antibody in Western Blot (WB) — Knockdown of RAC1, 2, 3 was achieved by separately transfecting HeLa cells with RAC1, 2, 3 specific siRNAs (Silencer® select Product # S11711, S11713 and S11715, S11716 and S11718, S11717 respectively). Western blot analysis (Fig. a) was performed using whole cell extracts from the untransfected cells (lane 1, 4, 7), non-targeting scrambled siRNA transfected cells (lane 2, 5, 8), RAC1 knockdown cells (lane 3), RAC2 knockdown cells (lane 6) and RAC3 knockdown cells (lane 9). The blot was probed with RAC1 Monoclonal Antibody (1301CT276.121.104) (Product # MA5-37658, 1:1,000) and Goat anti-Mouse IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177, 1:20,000). Densitometric analysis of this western blot is shown in histogram (Fig. b). Decrease in signal observed upon RAC1 specific siRNA mediated knock down confirms that antibody is specific to RAC1. An uncharacterized band (*) was observed at ~60 kDa.

RAC1 Antibody in Flow Cytometry (Flow) — Flow cytometry analysis performed on U-87 MG cells using RAC1 Monoclonal Antibody (Product # MA5-37658) at a 1:100 dilution compared to an isotype control of mouse IgG2b (blue). An Alexa Fluor® 488 goat anti-mouse lgG at 1:400 dilution was used as the secondary antibody.

Members of this superfamily appear to regulate a diverse array of cellular events, including the control of cell growth, cytoskeletal reorganization, and the activation of protein kinases. Two transcript variants encoding different isoforms have been found for this gene.
