SOX17 Antibody in Western Blot (WB) — Western blot was performed using Anti-SOX17 Polyclonal Antibody (Product # PA5-72815) and a 44 kDa band corresponding to Transcription factor SOX-17 was observed in iPSC differentiated to definitive endoderm and OVCAR-3 when compared to other cell lines. Nuclear enriched extracts (30 µg lysate) of iPSC (Lane 1), iPSC differentiated to definitive endoderm (Lane 2), COS-7 (Lane 3), Hep G2 (Lane 4), OVCAR-3 (Lane 5), IMR-32 (Lane 6), HUVEC (Lane 7) were electrophoresed using NuPAGE™ 10% Bis-Tris Protein Gel (Product # NP0301BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23002) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (0.5 µg/mL) and detected by chemiluminescence with Goat anti-Rabbit IgG (Heavy Chain) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:6000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).

SOX17 (transcription factor SOX-17) belongs to the SRY-related HMB box family of transcription factors. It acts as a transcription regulator that binds target promoter DNA and bends the DNA. It binds to the sequences 5'-AACAAT-'3 or 5'-AACAAAG-3'. SOX17 modulates transcriptional regulation via WNT3A and inhibits Wnt signaling. It promotes degradation of activated CTNNB1. SOX17 plays an essential role in the regulation of embryonic development and is required for normal looping of the embryonic hear tube and for normal development of the definitive gut endoderm. Mutations in the gene can result in vesicoureteral reflux.
