Organism Type | Mouse | T2 B cells were purified from splenic B cells by cell sorting. Splenic B cells were prepared as described previously. Then 5 x 107 splenic B cells were stained with FITC-conjugated anti-CD21, PE-conjugated anti-HSA, and biotinylated anti-CD23 combined with streptavidin-CyChrome. T2 B cells (CD21hiHSAhiCD23+B220+) were sorted. Staining with anti-CD21, anti-HSA, and anti-CD23 instead of anti-IgM and anti-IgD was performed to avoid contamination of marginal zone B cells and also in order to prevent potential activation signals through the BCR. |
