The blue is Hoechst staining of nuclear DNA. The ankyrin G antibody produces specific staining associated with the plasma membrane.

Ankyrin G Antibody in Immunocytochemistry (ICC/IF) — Immunofluorescent analysis of Ankyrin G in HEK293 cell culture. Samples were stained with Ankyrin G monoclonal antibody (Product # MA5-47397) using a dilution of 1:2,000 (green). The blue is Hoechst staining of nuclear DNA. The ankyrin G antibody produces specific staining associated with the plasma membrane.

Ankyrin G Antibody in Immunohistochemistry (PFA fixed) (IHC (PFA)) — Immunohistochemistry analysis of Ankyrin G in cortex section of rat brain. Samples were stained with Ankyrin G monoclonal antibody (Product # MA5-47397) using a dilution of 1:2,000 (green). Costained with rabbit pAb to NF-L dilution 1:5,000 in red. The blue is Hoechst staining of nuclear DNA. Following transcardial perfusion of rat with 4% paraformaldehyde, brain was post fixed for 24 hours, cut to 45 μM, and free-floating sections. The ankyrin G antibody stains the axonal initial segments, while the RPCA-NF-L antibody labels dendrites and axons of neuronal cells.

Ankyrin G Antibody in Western Blot (WB) — Western blot analysis of Ankyrin G in different tissue and cell lysates. Samples were incubated in Ankyrin G monoclonal antibody (Product # MA5-47397) using a dilution of 1:2,000 (green). [1] protein standard (red), [2] NIH-3T3, [3] C6, [4] HEK293, [5] HeLa, [6] SH-SY5Y cells, [7] rat brain, and [8] mouse brain lysates. Bands at ~190 kDa represent ankyrin3 splice variants, higher molecular weight bands at 270 kDa and 480 kDa can be seen on longer exposure. Lower molecular weight bands are likely proteolytic fragments.

Ankyrin-G is found in several tissues including brain where, due to tissue-specific RNA processing, it is expressed as two different isoforms, localized to axon initial segments and nodes of Ranvier. Ankyrin-G is also found at sites of epithelial cell-cell contact and in the intracellular membranes of some cells. Ankyrin-G isoforms contain a unique stretch of sequence highly enriched in serine and threonine residues immediately following a globular head domain. Ankyrin-G is thought to participate in the maintenance and targeting of ion channels and cell adhesion molecules to the nodes of Ranvier and to axon initial segments.
