Brd4 Antibody (ER1901-02) in ICC/IF
ICC staining of Brd4 in A549 cells (green). Formalin fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 10 minutes at room temperature and blocked with 10% negative goat serum for 15 minutes at room temperature. Cells were probed with the primary antibody (ER1901-02, 1:200) for 1 hour at room temperature, washed with PBS. Alexa Fluor®488 conjugate-Goat anti-Rabbit IgG was used as the secondary antibody at 1:1,000 dilution. The nuclear counter stain is DAPI (blue).

BRD4, a ubiquitously expressed nuclear phosphoprotein, is a member of BET protein family. It is a human homolog of mouse MCAP and consists of 2 N-terminal bromodomains, a central ET-domain, and several kinase-like motifs in its N-terminus. It is expressed as 2 isoforms; the longer variant has a C-terminus region with a praline and glutamine-rich domains. BRD4 is a chromatin-binding protein whose expression is induced in response to growth stimuli. BRD4 acts at different stages of the cell cycle by interacting with proteins including RFC and SPA-1 and regulates cell growth. It also has a role in regulating chromosomal dynamics during mitosis. BRD4 functions as a cellular target for E2 protein of papillomavirus.
