Cathepsin D Antibody in Immunocytochemistry (ICC/IF) — Immunofluorescence analysis of Cathepsin D was performed using 70% confluent log phase Neuro-2a cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 15 minutes, and blocked with 2% BSA for 45 minutes at room temperature. The cells were labeled with Cathepsin D Polyclonal Antibody (Product # PA5-79094) at 1:100 in 0.1% BSA, incubated at 4 degree celsius overnight and then labeled with Goat anti-Rabbit IgG (H+L) Highly Cross-Adsorbed Secondary Antibody, Alexa Fluor Plus 488 (Product # A32731), (1:2000), for 45 minutes at room temperature (Panel a: Green). Nuclei (Panel b:Blue) were stained with ProLong™ Diamond Antifade Mountant with DAPI (Product # P36962). F-actin (Panel c: Red) was stained with Rhodamine Phalloidin (Product # R415, 1:300). Panel d represents the merged image showing cytoplasmic localization. Panel e represents control cells with no primary antibody to assess background. The images were captured at 60X magnification.

Cathepsin D Antibody in Immunohistochemistry (Paraffin) (IHC (P)) — Immunohistochemical analysis of Cathepsin D in paraffin-embedded section of rat liver tissues. Heat mediated antigen retrieval was performed in citrate buffer (pH6, epitope retrieval solution) for 20 mins. The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 1μg/mL rabbit anti-Cathepsin D antibody (Product # PA5-79094) overnight at 4°C. Biotinylated goat anti-rabbit IgG was used as secondary antibody and incubated for 30 minutes at 37°C. The tissue section was developed using Strepavidin-Biotin-Complex (SABC) with DAB as the chromogen.

Cathepsin D Antibody in Western Blot (WB) — Western blot was performed using Anti-Cathepsin D Polyclonal Antibody (Product # PA5-79094) and a 42 kDa band corresponding to Cathepsin D was observed across cell lines and tissue extracts tested. Whole cell extracts (30 µg lysate) of 3T3-L1 (Lane 1), NIH/3T3 (Lane 2), RAW 264.7 (Lane 3), Neuro-2a (Lane 4) and tissue extracts of Mouse Liver (Lane 5), Mouse Testis (Lane 6), Mouse Heart (Lane 7) and Rat Liver (Lane 8) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (0.3 µg/mL) and detected by chemiluminescence with Goat anti-Rabbit IgG (Heavy Chain) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).

Cathepsin D Antibody in Western Blot (WB) — Western blot analysis of Cathepsin D in Lane 1: mouse liver tissue lysate, Lane 2: mouse brain tissue lysate, Lane 3: mouse thymus tissue lysate, Lane 4: NEURO whole cell lysate using 40-50 µg per well. Sample was incubated with Cathepsin D (Product # PA5-79094) at a dilution of 0.5 µg/mL.
