Immunofluorescence analysis of LRP6 was performed using 70% confluent log phase A549 cells. LRP6 Antibody in Immunocytochemistry (ICC/IF) — Immunofluorescence analysis of LRP6 was performed using 70% confluent log phase A549 cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.01% Triton™ X-100 for 15 minutes, and blocked with 2% BSA for 10 minutes at room temperature. The cells were labeled with LRP6 Polyclonal Antibody (Product # PA5-89161) at 1:100 dilution in 0.1% BSA, incubated at 4 degrees celsius overnight, and then labeled with Donkey anti-Rabbit IgG (H+L) Highly Cross-Adsorbed Secondary Antibody, Alexa Fluor Plus 488 (Product # A32790), (1:2000 dilution), for 45 minutes at room temperature (Panel a: Green). Nuclei (Panel b: Blue) were stained with Hoechst 33342 (Product # H1399). F-actin (Panel c: Red) was stained with Alexa Fluor™ Plus 647 Phalloidin (Product # A30107, 1:2000 dilution). Panel d represents the merged image showing endoplasmic reticulum localization. Panel e represents control cells with no primary antibody to assess the background. The images were captured at 40X magnification in CellInsight CX7 LZR High-Content Screening (HCS) Platform (Product # CX7A1110LZR).

LRP6 Antibody in Western Blot (WB) — Western blot was performed using Anti-LRP6 Polyclonal Antibody (Product # PA5-89161) and a 200-240 kDa band corresponding to LRP6 was observed across all cell lines tested except U-937 which expresses LRP6 at a relatively lower level. Nonspecific bands (*) were observed between 40-200 kDa. Whole-cell extracts (60 µg lysate) of A549 (Lane 1), HeLa (Lane 2), HAEC (Lane 3), and U-937 (Lane 4) were electrophoresed using NuPAGE™ 3-8% Tris-Acetate Protein Gel (Product # EA0378BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:500 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (Heavy Chain) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036,1:20,000 dilution) using the iBright™ FL1500 Imaging System (Product # A44115). Chemiluminescent detection was performed using SuperSignal™ West Atto Ultimate Sensitivity Substrate (Product # A38556).

LRP6 Antibody in Western Blot (WB) — Western blot analysis of LRP6 in extracts of Mouse liver. Samples were incubated with LRP6 Polyclonal antibody (Product # PA5-89161) using a dilution of 1:1,000, followed by HRP Goat Anti-Rabbit IgG (H+L) at a dilution of 1:10,000. Lysates/proteins: 25 µg per lane. Blocking buffer: 3% nonfat dry milk in TBST. Detection: ECL Basic Kit. Exposure time: 60s.

LRP6 is essential for the Wnt/beta catenin signaling pathway, probably by acting as a coreceptor together with Frizzled for Wnt. It is a specific, high-affinity receptor for DKK1 and DKK2, but not DKK3. The interaction with DKK1 blocks LRP6-mediated Wnt/beta catenin signaling via LRP6 removal via Kremen proteins-mediated endocytosis.
