RAF1/c-RAF Antibody (A301-519A) in WB
Detection of human and mouse RAF1/c-RAF by western blot (h&m) and immunoprecipitation (h). Samples: Whole cell lysate from HeLa (5, 15 and 50 µg for WB; 1 mg for IP, 20% of IP loaded), 293T (T; 50 µg) and mouse NIH3T3 (M; 50 µg)cells. Antibodies: Affinity purified rabbit anti-RAF1/c-RAF antibody A301-519A used for WB at 0.04 µg/ml (A) and 1 µg/ml (B) and used for IP at 3 µg/mg lysate. Raf-1 (c-Raf) is a MAP kinase kinase kinase (MAP3K) which functions downstream of the Ras family of membrane associated GTPases to which it binds directly. Once activated Raf-1 can phosphorylate to activate the dual specificity protein kinases MEK1 and MEK2 which in turn phosphorylate to activate the serine/threonine specific protein kinases ERK1 and ERK2. Activated ERKs are pleiotropic effectors of cell physiology and play an important role in the control of gene expression involved in the cell division cycle, apoptosis, cell differentiation and cell migration. Mutations in this gene are associated with Noonan syndrome 5 and LEOPARD syndrome 2.
