TIGIT Chimeric Antibody (MA5-48122) in ELISA
ELISA analysis of TIGIT chimeric monoclonal antibody (Product # MA5-48122) (red line) and isotype control (blue line) was performed by coating plate with TIGIT-Fc fusion protein at a concentration of 5 µg/mL. A 3-fold serial dilution from 10,000 to 0.17 ng/mL of the antibody was done. For signal detection, a 1:4,000 dilution of HRP-labelled anti-rabbit IgG1 antibody was used. The second increase in absorbance at 1,000 ng/mL is suggestive of a possible secondary binding epitope.

Specificity: 4D4 specifically recognises TIGIT.
Preparation: Armenian hamsters were immunized with recombinant mouse TIGIT tetramers by a combination of s.c. and food pad immunization and booster injections. Draining lymph nodes were fused with Sp2/0-Ag14 cells, selected in HAT (hypoxanthine/aminopterin/thymidine) medium and supernatants were screened for specificity by ELISA and flow cytometry using TIGIT-transfectants.
