The Human NOTCH-1 ELISA quantitates Hu NOTCH-1 in human serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu NOTCH-1.
原理方法
The Human NOTCH-1 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen.

The notch gene belongs to a family of epidermal growth factor (EGF) like homeotic genes, which encode transmembrane proteins with a variable number of cysteine-rich EGF-like repeats in the extracellular region. Four notch genes have been described in mammals: Notch1, Notch2, Notch3 and Notch4(Int-3), which have been implicated in the differentiation of the nervous system and other structures. The EGF-like proteins Delta and Serrate have been identified as ligands of Notch1. Mature Notch proteins are heterodimeric receptors derived from the cleavage of Notch pre-proteins into an extracellular subunit (NEC) containing multiple EGF-like repeats and a transmembrane subunit including intracellular region (Ntm).
