Applied Biosystems TaqMan MGB (minor groove binder) probes are dual-labeled probes used for real-time PCR applications using TaqMan chemistry. TaqMan MGB Probes incorporate a 5' fluorescent reporter dye and a 3' nonfluorescent quencher (NFQ). The NFQ offers the advantage of lower background signal, which results in better precision in quantitation. Custom TaqMan MGB probes are HPLC-purified and available with FAM, VIC, TET, and NED reporter dyes.
Shorter, more specific probesTaqMan MGB probes include a MGB moiety at the 3’ end that increases the melting temperature (Tm) of the probe and stabilizes probe/target hybrids. This means that TaqMan MGB probes can be significantly shorter than traditional probes, providing better sequence discrimination and flexibility to accommodate more targets.
Nonfluorescent quencher (NFQ) maximizes sensitivityTaqMan MGB probes incorporate an NFQ to absorb (quench) signal from the fluorescent dye label at the other end of the probe. The properties of the NFQ combined with the short length of the MGB probe result in lower background signal than with non-MGB NFQ probes. Lower background means increased sensitivity and precision in your data.
