LysoLight Antibody Labeling Kits provide a powerful tool for visualizing the lysosomal localization and catabolic degradation of antibodies and proteins. These kits utilize a unique fluorophore that is only active upon enzymatic cleavage by the lysosomal specific enzyme, Cathepsin B. Outside of the lysosome, LysoLight dyes have little to no observable background fluorescence, and only upon localization to the protease rich environment of the lysosome, does LysoLight become activated and brightly fluoresce bright green or deep red in color.
The labeling protocol provided in the LysoLight Antibody Labeling Kits involves a simple and rapid procedure, ensuring minimal hands-on time and reliable results. The kits include all necessary reagents, including the LysoLight dye, buffers, and purification columns, to facilitate the antibody labeling process. The LysoLight reactive dye contained in these kits readily reacts with lysine residues in antibodies or proteins to yield a covalently attached fluorogenic sensor. The spin columns included in the kits are used for purifying the labeled antibody from excess dye with yields of 70–95%. The kits contain sufficient reagents for three labeling reactions of 100 μg or 1 mg of antibody. The user guide describes a general protocol for using the amine-reactive form of the LysoLight dye for labeling either 100 μg or 1 mg of IgG.
